in BonnThe Neurobiology Laboratory at the Department of Neurology, University Bonn investigates the molecular, cellular and pharmacological aspects of the pathogenesis of neurodegenerative disorders with special emphasis on the cerebellar ataxias, Parkinson's disease and Multiple System Atrophy (MSA). Current research projects focus on the molecular mechanism of neuronal cell death in these disorders in comparison to normal aging and the pathophysiology of trinucleotide repeat disorders with particular interest in differentially regulated gene expression, gene expression profiling and epigentic mechanisms.
With a particular focus on spinocerebellar ataxia type 3 (SCA3), one of eight neurodegenerative disorders caused by an unstable CAG repeat expansion in the coding gene region, resulting in an expanded polyglutamine tract in the corresponding protein, ataxin-3, we analyze the molecular steps leading to neurodegeneration and the physiological protein functions. We have developed transgenic models using inducible high-level expression of non-expanded or expanded human full-length disease proteins, a AT3 knockout mouse and are currently resolving the physiological function of the respective proteins which are involved in key celullar mechanisms such as nucleocytoplasmic-shuttling, ER-stress and the ubiquitin proteasom system.
