Drosophila imaginal disc transdetermination, Polycomb & Trithorax function, chromatin, histone modifications

We investigate the genetic and epigenetic control of cellular determination in Drosophila imaginal disc development. Imaginal discs that give rise to the adult structures during metamorphosis are committed to their appropriate developmental program. Yet, by experimentally inducing regenerative growth these imaginal disc cells can be stimulated to change their developmental fate. This fate-switching system of imaginal disc transdetermination offers an entry point to analyze the underlying mechanisms at a genomic and molecular level. By microarray hybridization in combination with linear RNA amplification we identified chromatin regulators of the Polycomb- and Trithorax group (PcG, TrxG) that propagate a silenced or active state of transcription as potent regulators of transdetermination. To further investigate the underlying molecular mechanisms our group applies genetic and biochemical approaches to analyze the function of PcG factors in cellular determination.
